本书是在原版Encyclopedia of Toxicology(2nd edn)的基础上对词条进行的分类重排。编成三卷，分别为精选卷、拓展卷与社科管理卷，更加适合国内读者阅读。本书涵盖了毒理学研究的各个领域，从基础到前沿，甚至一些尚存争议的问题也包含其中。
本书收集了1057个条目，按英文字母顺序排列，内容详尽，信息量宽广。书中既有毒理学的基本理论、学科体系知识的介绍，也包含有毒理学专业知识、新的进展资料、毒理学派生出的相关社会科学问题(如历史事件、法律、条规、教育、国际和区域组织等)的信息资料。本百科全书还对近年来出现的毒理学新名词首次做了比较系统和权威性的解释，如鸟类生态毒理学、计算机毒理学、基准剂量、致癌效力因素、代谢组学等。很多在其他专著、教科书、工具书中查不到的毒理学或相关的信息资料，读者都能在此百科全书中找到。本百科全书在编写风格上也完全不同于以往出版的毒理学专著、教科书或工具书，真正做到了宽细结合、深浅恰当，适合于不同专业和层次的读者参阅。
第二版百科全书汇集了392作者提供的1057条目录，而第一版只包括200名作者提供的749条目录。实际上，第一版中所有条目的内容在第二版中都得到了充实，而某些条目则完全进行了更新。在第二版中新出现的308个条目中，包括鸟类生态毒理学、基准剂量、杀虫剂、计算机毒理学、致癌效力因素、代谢组学、化学事件、蒙地卡罗分析、非致死性化学武器、无脊椎动物生态毒理学、药物滥用、癌症化疗因子和消费产品等等。许多针对特殊化学物的条目都是全新的，而书中搜录的国际组织也得到了很大的扩展。某些条目罗列了许多著名的毒物中毒相关事件：如美国拉夫运河事件、泰晤士海滩事件、切尔诺贝利事件和三里岛事件等。另外许多条目涉及毒理学知识的社会应用，如文化毒理学、环境犯罪、著名投毒者和中毒可件、古代化学生物战以及美国环境运动历史等。因此，新版的毒理学百科全书在长度、广度、以及深度上都对毒理学的许多方面进行了更广泛和深入的概述。
